REGN2477 + REGN1033 is a combination therapy developed by Regeneron Pharmaceuticals for the treatment of sporadic inclusion body myositis (sIBM). The combination consists of two monoclonal antibodies: garetosmab (REGN2477), which targets activin A, and trevogrumab (REGN1033), which targets myostatin (GDF-8). Both activin A and myostatin are members of the TGF-beta superfamily that act as negative regulators of muscle mass. By blocking these ligands, the combination therapy aims to inhibit the myostatin/activin receptor signaling pathway, thereby promoting muscle growth and improving muscle function in patients with sIBM. A Phase 2 clinical trial was initiated to evaluate the efficacy of the combination on total lean mass, but Regeneron cancelled the study in late 2018.
