Overview

Garsorasib + cetuximab beta is a combination therapy being investigated for the treatment of KRAS G12C-mutated non-small cell lung cancer (NSCLC). Garsorasib (also known as D-1553 or goselasib) is a potent, selective, and covalent small molecule inhibitor that irreversibly binds to the GDP-bound state of the KRAS G12C mutant protein, locking it in an inactive conformation and preventing downstream oncogenic signaling. Cetuximab beta is a monoclonal antibody that targets the epidermal growth factor receptor (EGFR). The combination is designed to enhance therapeutic efficacy by simultaneously inhibiting the KRAS G12C mutation and blocking EGFR-mediated bypass signaling pathways, which are common mechanisms of resistance to KRAS G12C monotherapy. This combination is currently being evaluated in clinical trials led by Sun Yat-sen University Cancer Center.
