Overview

Gatipotuzumab + necitumumab is an investigational combination of two monoclonal antibodies: gatipotuzumab (targeting tumor-associated mucin 1, TA-MUC1) and necitumumab (targeting epidermal growth factor receptor, EGFR)[6][9]. - **Gatipotuzumab** is an anti-TA-MUC1 antibody, designed to bind the aberrantly glycosylated form of the mucin 1 protein commonly overexpressed in various epithelial cancers, inducing antibody-dependent cellular cytotoxicity and inhibiting tumor growth[6]. - **Necitumumab** is a human IgG1 monoclonal antibody that binds to the extracellular domain of EGFR, blocking ligand binding, dimerization, and downstream signaling, thereby inhibiting cell proliferation and survival and mediating antibody-dependent cellular cytotoxicity[2][5][7][8]. The combination aims to simultaneously inhibit two key cancer targets—TA-MUC1 and EGFR—potentially enhancing antitumor effects in solid tumors expressing EGFR and TA-MUC1. The therapeutic hypothesis is that dual targeting may help overcome resistance and improve efficacy in refractory patients[6][9]. Clinical studies (such as the GATTO trial) have tested this combination in advanced solid tumors with encouraging preliminary activity in patients progressing after standard therapy[6].
