**Gatipotuzumab + panitumumab** is a combination regimen of two monoclonal antibodies: gatipotuzumab, a glyco-engineered humanized antibody targeting the tumor-associated epitope of mucin-1 (TA-MUC1), and panitumumab, a human IgG2 monoclonal antibody that binds the extracellular domain of the epidermal growth factor receptor (EGFR), preventing its activation. Gatipotuzumab facilitates immune-mediated tumor cell killing by targeting TA-MUC1, while panitumumab inhibits EGFR-driven signaling in epithelial tumors. This dual targeting is designed to exploit the synergy between TA-MUC1 and EGFR in driving carcinogenesis, aiming for enhanced anti-tumor activity in patients with refractory solid tumors expressing both antigens. The combination has been explored in metastatic colorectal cancer (mCRC) and other solid tumors in the phase Ib GATTO study[3][5][7].
