GBM6-AD + poly-ICLC is a cancer vaccine combining tumor-cell lysate (GBM6-AD) with poly-ICLC as an immune adjuvant. The GBM6-AD component is derived from the GBM6 cell line, which expresses antigens commonly found in glioma cells. The preparation involves culturing tumor cells, lysing them to release tumor-associated antigens (TAAs), and combining this lysate with poly-ICLC. Poly-ICLC (Polyinosinic-Polycytidylic Acid Stabilized with Polylysine and Carboxymethylcellulose) acts as a potent immune adjuvant by activating pattern recognition receptors like toll-like receptor 3 (TLR3) on dendritic cells, leading to production of type-1 interferons and cytokines that promote a Th1-type immune response. The vaccine is administered intradermally to effectively deliver it to immune cells in the skin, particularly dendritic cells, which then present the antigens to T cells in the lymph nodes.
