Overview

GC1118 + FOLFIRI is an experimental combination regimen consisting of **GC1118**, a novel anti-epidermal growth factor receptor (EGFR) monoclonal antibody, and **FOLFIRI**, a standard chemotherapy regimen containing irinotecan, leucovorin (folinic acid), and 5-fluorouracil. GC1118 acts by inhibiting EGFR with enhanced blockade of both low- and high-affinity EGFR ligands, and has demonstrated antitumor activity and safety in early-phase studies. FOLFIRI works by disrupting DNA synthesis and function—irinotecan inhibits topoisomerase I, 5-fluorouracil is an antimetabolite interfering with RNA and DNA, and leucovorin enhances 5-FU action. This combination is being evaluated as a second-line treatment for RAS and BRAF wild-type recurrent or metastatic colorectal cancer and is currently in phase 1b/2a studies[1][3][5].
