Overview

GC1118 + irinotecan is an investigational combination therapy evaluated primarily as a treatment for metastatic colorectal cancer, especially in patients with KRAS/NRAS/BRAF wild-type tumors resistant to first-line therapy. GC1118 is a novel fully human monoclonal antibody targeting the epidermal growth factor receptor (EGFR) with high blocking activity against both high- and low-affinity EGFR ligands. Irinotecan is a small molecule chemotherapy agent classified as a DNA topoisomerase I inhibitor, which prevents DNA repair in rapidly dividing cancer cells, leading to cell death. The combination has been studied in phase 1b/2a clinical trials, with GC1118 administered weekly (3 mg/kg) and irinotecan biweekly (180 mg/m^2) either alone or as part of the FOLFIRI regimen. This regimen aims to leverage both targeted EGFR inhibition and cytotoxic chemotherapy to improve tumor response in advanced, refractory cases[1][3][5][7].
