Overview

GC33 + sorafenib is a combination therapy of **codrituzumab** (also known as GC33 or RO5137382), a recombinant, fully humanized monoclonal antibody targeting glypican-3 (GPC3), and **sorafenib**, a small molecule multikinase inhibitor. GC33 binds to GPC3, an oncofetal protein overexpressed in hepatocellular carcinoma (HCC), mediating antibody-dependent cellular cytotoxicity (ADCC), while sorafenib targets multiple kinases involved in tumor cell proliferation and angiogenesis (including VEGFR2, PDGFR, RAF kinases). The combination has been evaluated and shown to have an additive antitumor effect in preclinical models and clinical studies in patients with advanced HCC. Clinical trials report that the regimen is generally tolerable, with expected toxicities, but did not achieve superior efficacy compared to single-agent sorafenib[1][2].
