Overview

Gd(DOTA-FPbG) is a tissue-specific, gadolinium-based magnetic resonance imaging (MRI) contrast agent designed to target cells expressing the enzyme β-glucuronidase (βG). It consists of a gadolinium-DOTA (gadoterate) chelate conjugated to a β-glucuronide moiety via a fluoropropyl linker. This agent functions as an activatable or 'smart' probe; the presence of β-glucuronidase, which is often overexpressed in certain tumors and the necrotic tumor microenvironment, triggers the cleavage of the glucuronide group. This enzymatic activation results in a change in the agent's pharmacokinetic properties or relaxivity, leading to significantly enhanced T1-weighted signal intensity specifically within βG-positive tissues. Preclinical studies in murine liver tumor models have shown that Gd(DOTA-FPbG) provides superior contrast enhancement and tumor-to-liver differentiation compared to conventional non-specific agents like Gd-DOTA.
