Gd-DTPA + DACHPt refers to a drug delivery system in which polymeric micelles are loaded with both gadolinium-diethylenetriaminopentaacetic acid (Gd-DTPA), an MRI contrast agent, and (1,2-diaminocyclohexane)platinum(II) (DACHPt), the parent complex of the anticancer drug oxaliplatin[1]. This combination enables simultaneous imaging (MRI-detectable) and therapy (chemotherapy) of tumors. In studies with orthotopic rat models of hepatocellular carcinoma, Gd-DTPA/DACHPt-loaded micelles injected via the hepatic artery provided strong and specific contrast enhancement for tumor detection and induced high levels of tumor apoptosis, significantly reducing tumor size and improving survival[1]. The mechanism involves tumor targeting via the enhanced permeability and retention (EPR) effect of nanomicelles, release of the cytotoxic platinum agent for antitumor activity, and Gd-DTPA for MRI visualization. This system is a promising approach for theranostic applications in advanced hepatocellular carcinoma.
