The **gD vaccine + 3D-MPL** is a combination vaccine formulation that consists of a herpes simplex virus glycoprotein D antigen (gD, often expressed as gD 2t or similar glycoprotein constructs) and the adjuvant 3-O-deacylated monophosphoryl lipid A (3D-MPL). 3D-MPL functions as a Toll-like receptor 4 (TLR4) agonist, leading to strong stimulation of innate immunity, preferential induction of Th1-type immune responses, and enhanced production of IgG2a. When combined with gD antigen in a vaccine, 3D-MPL boosts both cell-mediated and humoral immune responses, critically enhancing cytolytic T lymphocyte (CTL) induction and overall vaccine immunogenicity. Research indicates this combination is especially associated with increased protection against pathogens where strong Th1/CTL responses are desirable, such as herpesviruses and HIV. The combination is often formulated with an oil-in-water emulsion for further adjuvant synergy and formulation stability[2][5].
