GD2-SADA:177Lu-DOTA is a two-step radioimmunotherapeutic drug complex under investigation for the treatment of solid tumors known to express GD2, including small cell lung cancer, high-risk neuroblastoma, sarcoma, and malignant melanoma. The therapy utilizes a bispecific fusion protein (GD2-SADA) that targets the tumor-associated antigen GD2 and serves as a platform for site-specific delivery of the radioactive payload, 177Lu-DOTA (Lutetium-177 chelated to DOTA). In the stepwise protocol, GD2-SADA is first administered to bind selectively to GD2-expressing tumor cells. Subsequent administration of 177Lu-DOTA delivers localized irradiation directly to the tumor site by binding to pretargeted GD2-SADA, thereby minimizing off-target toxicity. GD2-SADA rapidly clears from the bloodstream, increasing targeting specificity and reducing radiation exposure to healthy tissue. This drug represents a novel approach to pre-targeted radioimmunotherapy, leveraging the SADA technology developed at Memorial Sloan Kettering and licensed to Y-mAbs Therapeutics[4][6][7][8].
