Overview

This is a **four-drug combination regimen** consisting of GDC-0980 (a selective PI3K/mTOR inhibitor), bevacizumab (a recombinant humanized monoclonal antibody targeting vascular endothelial growth factor A), paclitaxel (a taxane-class microtubule inhibitor), and carboplatin (a platinum-based chemotherapeutic agent). GDC-0980 acts by inhibiting both PI3K and mTOR pathways, potentially overcoming resistance mechanisms in cancer cells. Bevacizumab blocks angiogenesis by neutralizing VEGF-A, impeding tumor vascularization. Paclitaxel disrupts mitosis by stabilizing microtubules, thereby preventing cell division, while carboplatin generates DNA cross-links, promoting apoptosis. This combination is under investigation for the treatment of solid tumors, particularly ovarian and lung cancers, aiming to synergize cytotoxic, anti-angiogenic, and pathway-targeted effects.
