Gedatolisib + trastuzumab is a combination therapy that targets both the PI3K/mTOR signaling pathway and HER2 receptor in cancer cells. Gedatolisib is a dual PI3K/mTOR inhibitor that blocks the mTOR/PIK3 signaling pathway involved in tumor growth and proliferation. Trastuzumab is a monoclonal antibody that binds to the HER2/neu receptor, reducing HER2 expression and inhibiting angiogenesis that supports tumor growth. This combination has shown particular promise in patients with HER2-positive metastatic breast cancer with PIK3CA aberrations who have progressed after multiple lines of therapy.
