Overview

Gefitinib + bevacizumab is a combination therapy consisting of two distinct agents used primarily in the treatment of advanced non-small cell lung cancer (NSCLC) with epidermal growth factor receptor (EGFR) mutations. Gefitinib is an oral small molecule tyrosine kinase inhibitor that selectively inhibits the EGFR tyrosine kinase, thereby blocking downstream signaling pathways involved in tumor cell proliferation and survival. Bevacizumab is a recombinant humanized monoclonal antibody targeting vascular endothelial growth factor A (VEGF-A), inhibiting angiogenesis by preventing VEGF from binding to its receptors on endothelial cells, thus reducing tumor blood supply and growth. The combination aims to simultaneously inhibit both tumor cell proliferation and angiogenesis, potentially overcoming resistance mechanisms seen with EGFR inhibitors alone and improving progression-free survival in patients with EGFR-mutant NSCLC[2][3][7].
