Overview

gefitinib + capecitabine + oxaliplatin is a combination chemotherapy and targeted therapy regimen investigated for the treatment of metastatic colorectal cancer. This combination, studied in a Phase I/II trial led by the Roswell Park Cancer Institute, pairs the EGFR-targeted tyrosine kinase inhibitor gefitinib (Iressa) with the XELOX regimen, which consists of the oral fluoropyrimidine capecitabine (Xeloda) and the intravenous platinum agent oxaliplatin (Eloxatin). The therapeutic strategy aimed to exploit the synergistic potential of blocking EGFR-mediated survival signaling while simultaneously inducing DNA damage and inhibiting nucleotide synthesis in tumor cells. Although preclinical models suggested enhanced activity, the clinical development of this specific triplet for colorectal cancer did not proceed to late-stage trials.
