These small molecules function by competitively binding to the adenosine triphosphate (ATP) binding site of the EGFR tyrosine kinase domain, thereby inhibiting the autophosphorylation of the receptor and blocking downstream signaling pathways such as PI3K/Akt, MAPK/ERK, and JAK/STAT that promote tumor cell growth and survival. The SINDAS trial specifically investigated the efficacy of these TKIs when combined with consolidative stereotactic body radiation therapy (SBRT) compared to TKI monotherapy, demonstrating improved progression-free and overall survival in the combination arm.
