Gefitinib + pembrolizumab is a combination therapy that pairs gefitinib, an epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor, with pembrolizumab, an anti-programmed death 1 (PD-1) immunotherapy antibody. This combination was evaluated in the phase 1/2 KEYNOTE-021 study for patients with previously untreated stage IIIB/IV EGFR-mutant non-small cell lung cancer (NSCLC). However, the combination was found not to be feasible due to significant liver toxicity, with 5 out of 7 patients (71.4%) developing grade 3/4 liver toxicity, leading to permanent treatment discontinuation in 4 patients.
