Overview

This is a combination therapy consisting of three agents: - **Gemcitabine** is a nucleoside analog used as chemotherapy that inhibits DNA synthesis, leading to cell death in rapidly dividing tumor cells. - **Apatinib** (sometimes misspelled as "aptinib") is a small molecule tyrosine kinase inhibitor targeting vascular endothelial growth factor receptor 2 (VEGFR2), thereby inhibiting angiogenesis and reducing blood supply to tumors. - **Toripalimab** is a humanized monoclonal antibody that targets programmed cell death protein 1 (PD-1) on T cells, functioning as an immune checkpoint inhibitor to enhance anti-tumor immune responses. The triple combination has been investigated primarily for recurrent or metastatic nasopharyngeal carcinoma (RM-NPC). Early-phase clinical trials suggest this regimen demonstrates promising antitumor activity and manageable toxicity. The combination leverages the cytotoxic effects of chemotherapy, antiangiogenic blockade, and immunotherapy for potential synergistic efficacy[1][3][5].
