Overview

This refers to an investigational treatment regimen developed at Memorial Sloan Kettering Cancer Center (MSKCC) involving the intraperitoneal (IP) administration of gemcitabine in combination with IP cisplatin. Gemcitabine is a pyrimidine nucleoside analog that interferes with DNA synthesis by inhibiting ribonucleotide reductase and competing with deoxycytidine triphosphate for incorporation into DNA. Cisplatin is a platinum-based chemotherapeutic agent that forms covalent bonds with DNA bases, resulting in intra-strand and inter-strand cross-links that inhibit DNA replication and transcription. The regimen was specifically studied in Phase I/II trials for patients with refractory or recurrent epithelial ovarian cancer, fallopian tube cancer, or primary peritoneal cancer who had persistent disease after standard platinum-based therapy. The protocol was designed to deliver high local concentrations of chemotherapy directly into the peritoneal cavity while minimizing systemic toxicity.
