The combination of gemcitabine, cisplatin, docetaxel, and fluorouracil refers to the cytotoxic chemotherapy agents used in the GP (gemcitabine and cisplatin) and TPF (docetaxel, cisplatin, and fluorouracil) induction regimens. These regimens have been evaluated in Phase 3 clinical trials, notably by West China Hospital, as induction chemotherapy for patients with locoregionally advanced nasopharyngeal carcinoma (LA-NPC). Gemcitabine and fluorouracil are antimetabolites that interfere with DNA synthesis by inhibiting ribonucleoside reductase and thymidylate synthase, respectively. Cisplatin is a platinum-based agent that induces DNA cross-linking, while docetaxel is a taxane that stabilizes microtubules to inhibit mitotic progression. These agents are typically used as neoadjuvant therapy prior to radiotherapy or concurrent chemoradiotherapy to reduce tumor burden and improve survival outcomes in advanced disease settings.
