Overview

This combination treatment regimen consists of the chemotherapy agents gemcitabine and cisplatin, and the immune checkpoint inhibitor durvalumab. It is being evaluated by Inova Health Care Services as an induction therapy for locally advanced unresectable intrahepatic cholangiocarcinoma. Gemcitabine and cisplatin are cytotoxic small molecules that inhibit DNA synthesis and cause DNA cross-linking, respectively. Durvalumab is a human monoclonal antibody that targets programmed death-ligand 1 (PD-L1), preventing it from binding to PD-1 and CD80, thereby enhancing the T-cell-mediated anti-tumor immune response. In the clinical trial context, this triplet is followed by Yttrium-90 (Y-90) radioembolization and consolidation therapy.
