Drug intelligence / Profile preview
Gemcitabine + cisplatin + immune checkpoint inhibitor (GemCis-ICI) is a combination chemoimmunotherapy regimen used in the treatment of advanced malignancies, most notably biliary tract cancer and urothelial carcinoma. The regimen combines two cytotoxic chemotherapeutic agents—gemcitabine (a nucleoside analog that inhibits DNA synthesis) and cisplatin (a platinum-based agent that cross-links DNA)—with an immune checkpoint inhibitor (a monoclonal antibody targeting the PD-1/PD-L1 pathway, such as durvalumab, pembrolizumab, or nivolumab). By pairing the immunogenic cell death induced by chemotherapy with the restoration of T-cell antitumor activity via checkpoint blockade, the regimen achieves synergistic therapeutic efficacy. It is established as a standard first-line treatment for unresectable or metastatic biliary tract cancer and advanced urothelial carcinoma.
