Gemcitabine + cisplatin + nab-paclitaxel + tislelizumab is an investigational four-drug combination regimen composed of three cytotoxic chemotherapies—gemcitabine (a nucleoside analog), cisplatin (a platinum-based DNA crosslinker), and nanoparticle albumin-bound paclitaxel (nab-paclitaxel, a microtubule inhibitor)—plus tislelizumab, a monoclonal antibody immune checkpoint inhibitor targeting PD-1. This combination is being explored for the treatment of advanced or metastatic solid tumors, particularly biliary tract cancers and potentially other gastrointestinal malignancies. The chemotherapy agents act synergistically to induce cancer cell death through DNA damage and mitotic arrest, while tislelizumab enhances anti-tumor immunity by blocking PD-1-mediated immunosuppression on T cells[1][4][5][10]. Tislelizumab has been approved in combination with platinum-based chemotherapy for certain gastric and esophageal cancers[6][8], but its use with this specific triplet chemotherapy backbone remains investigational.
