Gemcitabine + cisplatin + oxaliplatin is an investigational combination chemotherapy regimen composed of three cytotoxic agents—gemcitabine, cisplatin, and oxaliplatin. Each drug has a distinct mechanism of action targeting DNA synthesis or function. Gemcitabine is a nucleoside analog that inhibits DNA synthesis by incorporating into DNA and causing chain termination. Cisplatin and oxaliplatin are platinum-based compounds that form DNA crosslinks, leading to apoptosis in rapidly dividing cells. This triple-drug combination is not a standard regimen but may be explored in clinical trials for cancers where doublet regimens (such as gemcitabine with either cisplatin or oxaliplatin) have shown efficacy, including advanced biliary tract cancer and pancreatic cancer[1][6]. The rationale for combining all three drugs would be to potentially enhance antitumor activity by leveraging their complementary mechanisms; however, this approach may also increase toxicity.
