This is an investigational chemo-immunotherapy combination regimen consisting of the antimetabolite gemcitabine, the platinum-based alkylating agent cisplatin, and the anti-PD-1 monoclonal antibody toripalimab. It is primarily being evaluated by Sun Yat-sen University for the treatment of locally advanced nasopharyngeal carcinoma (NPC). In this regimen, gemcitabine and cisplatin (GP) serve as induction chemotherapy to reduce tumor burden and potentially release neoantigens, while toripalimab (developed by Shanghai Junshi Biosciences) blocks the interaction between PD-1 on T-cells and its ligands (PD-L1/PD-L2) on tumor cells, thereby enhancing the immune system's ability to identify and destroy cancer cells. This combination is typically administered as induction therapy followed by concurrent chemoradiotherapy (CCRT).
