Gemcitabine + cytarabine is a combination chemotherapy regimen consisting of two nucleoside analog antimetabolites used primarily in the treatment of hematologic malignancies such as leukemias. Both drugs are incorporated into DNA during replication, leading to inhibition of DNA chain elongation and ultimately causing apoptosis of malignant cells. Gemcitabine (a fluorinated cytidine analog) has a broader spectrum of antitumor activity compared to cytarabine but shares a similar mechanism involving incorporation into DNA and blocking synthesis. The combination has shown synergistic effects in preclinical models, enhancing the activity against leukemia cells by increasing cytotoxicity through complementary mechanisms targeting DNA synthesis[1][2]. This regimen is investigational for various leukemias and other cancers.
