Overview

Gemcitabine plus dacarbazine is a chemotherapy combination regimen used primarily for the treatment of advanced soft tissue sarcomas (STS) in patients who have failed, are unsuitable for, or are intolerant of treatment with anthracyclines or ifosfamide. This combination has demonstrated superior efficacy compared to dacarbazine alone in terms of progression-free survival (4.2 vs 2 months) and overall survival (16.8 vs 8.2 months) in randomized phase II trials. Gemcitabine is a nucleoside metabolic inhibitor that works by replacing building blocks of nucleic acids during DNA elongation, while dacarbazine is an alkylating agent. The combination is administered intravenously, with gemcitabine given as a fixed-dose rate infusion followed by dacarbazine.
