Gemcitabine + dexamethasone + cisplatin, commonly referred to as the GDP regimen, is a combination chemotherapy protocol used primarily as salvage therapy for relapsed or refractory Hodgkin lymphoma and other lymphomas. Gemcitabine is a nucleoside analog that inhibits DNA synthesis by incorporating into DNA and causing chain termination. Cisplatin is a platinum-based chemotherapeutic agent that forms DNA crosslinks, leading to apoptosis. Dexamethasone is a synthetic glucocorticoid with anti-inflammatory and immunosuppressive properties; in this regimen, it helps reduce inflammation and manage side effects of chemotherapy. The GDP regimen has demonstrated efficacy in inducing remission prior to autologous stem cell transplantation and is generally well-tolerated with manageable toxicity profiles[1][3][5].
