The combination of gemcitabine, docetaxel, and capecitabine (GTX) is a multi-drug chemotherapy regimen primarily used in the treatment of advanced pancreatic cancer. Gemcitabine is a nucleoside analog that inhibits DNA synthesis. After uptake into cancer cells, it is phosphorylated to form active metabolites that compete with deoxycytidine triphosphate for incorporation into DNA, leading to DNA fragmentation and apoptosis. It also inhibits ribonucleotide reductase. When combined with docetaxel and capecitabine in a specific sequence, this regimen can induce a p53-independent pathway of cell death through the MAP kinase pathway.
