Overview

This is a four-drug chemotherapy combination consisting of gemcitabine, docetaxel, irinotecan (also known as CPT-11), and cisplatin. Each component is a cytotoxic agent with distinct mechanisms of action: - **Gemcitabine** is a nucleoside analog that inhibits DNA synthesis by incorporation into DNA and inhibition of ribonucleotide reductase. - **Docetaxel** is a taxane that promotes microtubule assembly while inhibiting their disassembly, leading to cell cycle arrest. - **Irinotecan (CPT-11)** is a topoisomerase I inhibitor that prevents DNA unwinding necessary for replication and transcription. - **Cisplatin** forms DNA crosslinks, interfering with DNA repair and replication. These agents are often used in various combinations for the treatment of solid tumors such as non-small cell lung cancer (NSCLC) and other advanced malignancies. The rationale for combining these drugs lies in their complementary mechanisms of action and potential synergistic effects against tumor cells[1][2][3][4].
