Overview

This is a combination therapy consisting of three agents: - Gemcitabine, a nucleoside analog that inhibits DNA synthesis and is used as an antimetabolite chemotherapeutic. - Erlotinib, a small molecule inhibitor of the epidermal growth factor receptor (EGFR) tyrosine kinase. - Linsitinib (OSI-906), an oral small molecule dual inhibitor targeting both the insulin-like growth factor 1 receptor (IGF-1R) and the insulin receptor. This combination has been studied in clinical trials for metastatic ductal adenocarcinoma of the pancreas. The rationale for combining these agents is to target multiple pathways involved in tumor proliferation and survival, potentially overcoming resistance mechanisms seen with single-agent therapies[1][2][4].
