gemcitabine + fluorouracil + aldesleukin + interferon alfa is a combination chemobiotherapy regimen investigated by the Cancer Biotherapy Research Group for the treatment of metastatic or unresectable renal cell carcinoma. This regimen integrates cytotoxic chemotherapy with biological response modifiers to achieve a synergistic antitumor effect. Gemcitabine and fluorouracil are antimetabolites that interfere with DNA synthesis and repair, leading to cell cycle arrest and apoptosis in rapidly dividing cancer cells. Aldesleukin (recombinant interleukin-2) and interferon alfa are cytokines that modulate the host immune system, enhancing the proliferation and activity of cytotoxic T-cells and natural killer (NK) cells against tumor targets. By combining direct cytotoxicity with immune stimulation, the regimen aims to overcome the resistance often observed in advanced renal cell carcinoma.
