Overview

This combination therapy consists of the chemotherapy agents gemcitabine and nab-paclitaxel (the AG regimen) administered alongside two immune checkpoint inhibitors: iparomlimab (AK102), an anti-CTLA-4 monoclonal antibody, and tuvorilimab (AK127), an anti-TIGIT monoclonal antibody. The regimen is being evaluated in a Phase II clinical trial sponsored by Xinhua Hospital (Shanghai Jiao Tong University School of Medicine) for the treatment of advanced unresectable gallbladder cancer. The combination aims to enhance anti-tumor immune responses by simultaneously targeting the CTLA-4 and TIGIT pathways while utilizing the cytotoxic effects of chemotherapy to promote immunogenic cell death and improve surgical conversion rates. Both iparomlimab and tuvorilimab were developed by Akeso Bio.
