gemcitabine + nab-paclitaxel + metformin + multi-ingredient dietary supplement is a combination therapeutic regimen developed by the City of Hope Medical Center for the treatment of unresectable pancreatic adenocarcinoma. This regimen integrates standard cytotoxic chemotherapy (gemcitabine and nab-paclitaxel) with the metabolic modulator metformin and a complex multi-ingredient dietary supplement (DS) containing various vitamins, minerals, and phytochemicals like curcumin and EGCG. Gemcitabine and nab-paclitaxel work synergistically to inhibit DNA synthesis and stabilize microtubules, respectively. Metformin, an AMPK activator, is included to target the metabolic vulnerabilities of pancreatic cancer cells, potentially overcoming chemoresistance. The dietary supplement is intended to provide broad-spectrum inhibition of multiple oncogenic signaling pathways and reduce systemic inflammation. This combination is currently being evaluated in Phase 1 clinical trials to determine safety and feasibility.
