Overview

This is a combination regimen of three drugs used in the treatment of cancer, particularly pancreatic adenocarcinoma. Gemcitabine is a nucleoside analog that inhibits DNA synthesis, leading to cell death in rapidly dividing cells. Nab-paclitaxel (albumin-bound paclitaxel) stabilizes microtubules and prevents their disassembly, inhibiting mitosis. Vismodegib (GDC-0449) is an oral small-molecule inhibitor of the Hedgehog signaling pathway by antagonizing Smoothened (SMO), thereby blocking downstream signaling involved in tumor growth and maintenance of cancer stem cells. This combination has been investigated for synergistic effects against pancreatic cancer, with studies showing enhanced inhibition of tumor growth compared to standard regimens[1][2].
