Overview

Gemcitabine + oxaliplatin + dexamethasone is a combination chemotherapy regimen used primarily in the treatment of relapsed or refractory lymphomas, including aggressive B-cell non-Hodgkin lymphoma and peripheral T-cell lymphomas. - **Gemcitabine** is a nucleoside analog that inhibits DNA synthesis by incorporating into DNA strands and inhibiting ribonucleotide reductase, leading to cell cycle arrest and apoptosis. - **Oxaliplatin** is a platinum-based chemotherapeutic agent that forms DNA crosslinks, disrupting DNA replication and transcription, ultimately inducing cell death. - **Dexamethasone** is a synthetic glucocorticoid corticosteroid with anti-inflammatory and immunosuppressive properties; in oncology regimens it helps reduce inflammation, manage side effects of chemotherapy, and may have direct antitumor effects in certain hematologic malignancies. This combination (sometimes referred to as "GemDOx") has been studied as an alternative salvage therapy for patients who are not candidates for high-dose chemotherapy or stem cell transplantation. It has demonstrated efficacy with manageable toxicity profiles in phase II studies for relapsed/refractory aggressive lymphomas[1][3]. The regimen leverages the synergistic cytotoxicity of gemcitabine and oxaliplatin along with the supportive role of dexamethasone.
