Gemcitabine + paclitaxel is a combination chemotherapy regimen used primarily for the treatment of advanced or metastatic breast cancer after failure of prior anthracycline-containing adjuvant chemotherapy, unless anthracyclines were clinically contraindicated. It is also used in the management of advanced bladder cancer. The combination may be referred to as "GemTaxol." - **Mechanism of Action:** Both drugs are cytotoxic agents that target rapidly dividing cells. - *Gemcitabine* is a nucleoside analog (antimetabolite) that is phosphorylated intracellularly to active metabolites which inhibit DNA synthesis by incorporating into DNA and causing masked chain termination, leading to apoptosis in malignant cells. - *Paclitaxel* stabilizes microtubules and prevents their depolymerization during cell division, thereby inhibiting mitosis and promoting cell death[1][4][5]. - **Administration:** Both drugs are administered intravenously in cycles; typical regimens involve infusions every three weeks for up to six cycles[1].
