Overview

A multi-agent anticancer regimen combining the antimetabolite nucleoside analog gemcitabine, the microtubule-stabilizing taxane paclitaxel, and the anti-angiogenic monoclonal antibody bevacizumab. Gemcitabine inhibits DNA synthesis via intracellular conversion to active nucleotides (dFdCDP/dFdCTP) and ribonucleotide reductase inhibition, paclitaxel stabilizes microtubules to arrest mitosis and induce apoptosis, and bevacizumab binds VEGF-A to block VEGF receptor signaling and tumor angiogenesis. Phase II studies have evaluated this triplet as first-line therapy in HER2-negative metastatic breast cancer, reporting high response rates and progression-free survival around 12 months with manageable toxicity profiles[4][1][10]. Gemcitabine plus paclitaxel is an established combination for metastatic breast cancer; bevacizumab is an approved anti-VEGF therapy used with chemotherapy in several cancers[5][6][3][9].
