Overview

Gemcitabine + PM060184 is an investigational combination therapy for advanced solid tumors. Gemcitabine is a nucleoside analog and antimetabolite chemotherapeutic agent that inhibits DNA synthesis by incorporation into DNA and inhibition of ribonucleotide reductase, leading to cell cycle arrest and apoptosis. It is approved for use in various cancers including ovarian cancer, non-small cell lung cancer, breast cancer, pancreatic cancer, and bladder cancer[1][2][5][6]. PM060184 (also known as plocabulin) is a novel tubulin inhibitor developed by PharmaMar. It binds to tubulin with high affinity at a unique site distinct from other tubulin-binding agents, disrupting microtubule dynamics essential for mitosis and cellular transport. The combination aims to inhibit both DNA replication (via gemcitabine) and microtubule function (via PM060184), potentially providing synergistic anti-tumor activity[3][4][7]. The combination has been studied in phase 1 clinical trials for patients with advanced solid tumors including gastrointestinal stromal tumor (GIST), particularly those resistant to standard therapies such as imatinib[3][4].
