Overview

Gemcitabine + raltitrexed is a combination chemotherapy regimen used primarily in the treatment of advanced pancreatic cancer. Gemcitabine is a nucleoside analog that inhibits DNA synthesis by incorporating into DNA and causing chain termination, leading to apoptosis of rapidly dividing tumor cells. Raltitrexed is a thymidylate synthase inhibitor from the antimetabolite class, which blocks the synthesis of thymidine required for DNA replication. The combination has been studied in phase II clinical trials and demonstrated moderate efficacy with manageable toxicity profiles, offering an alternative to other regimens such as continuous-infusion 5-fluorouracil[1][2]. This regimen is considered convenient due to its administration schedule and has shown activity in patients with advanced or metastatic pancreatic adenocarcinoma[1][2].
