Gemcitabine + trabectedin is a combination chemotherapy regimen that has been investigated for the treatment of advanced solid tumors, particularly soft tissue sarcomas. Gemcitabine is a nucleoside analog that works by replacing the building blocks of nucleic acids during DNA elongation, arresting tumor growth and promoting apoptosis of malignant cells[3]. It is FDA-approved for various cancers including pancreatic cancer, non-small cell lung cancer, breast cancer, and ovarian cancer[3]. Trabectedin is an alkylating agent that is indicated for the treatment of patients with unresectable or metastatic liposarcoma or leiomyosarcoma who received prior therapy with an anthracycline-containing regimen[5]. The combination was studied to determine if the different mechanisms of action could provide synergistic effects against various solid tumors, particularly L-sarcomas (liposarcoma and leiomyosarcoma)[1][6].
