Gemcitabine + treosulfan is a combination chemotherapy regimen consisting of gemcitabine, a nucleoside analog that inhibits DNA synthesis, and treosulfan, an alkylating agent that induces DNA crosslinking. The combination has demonstrated synergistic cytotoxic effects in vitro and in clinical studies across several tumor types. Mechanistically, gemcitabine causes the persistence of treosulfan-induced DNA interstrand crosslinks, enhancing antitumor activity. This regimen has been investigated primarily for metastatic uveal melanoma and platinum-resistant ovarian cancer but also shows potential activity against pancreatic cancer cell lines. Clinical trials have reported disease stabilization and partial responses in these indications[1][2][3][4].
