gemcitabine + vincristine + cisplatin

The GVP regimen is a combination chemotherapy protocol consisting of gemcitabine, vincristine, and cisplatin, specifically investigated by Fudan University Shanghai Cancer Center for the treatment of advanced soft tissue sarcoma. Gemcitabine acts as a nucleoside analog that inhibits ribonucleoside reductase and terminates DNA chain elongation. Vincristine is a vinca alkaloid that binds to tubulin, inhibiting microtubule assembly and causing mitotic arrest. Cisplatin is a platinum-based agent that forms inter- and intra-strand DNA cross-links, interfering with DNA replication and transcription. This three-drug combination was evaluated in a Phase II clinical trial (NCT01192633) as a second-line therapy for patients with locally advanced or metastatic sarcoma who had failed prior anthracycline-based treatments.
