**GEN-001 + avelumab** is a combination therapy comprising a live biotherapeutic product, GEN-001—a single-strain live lyophilized *Lactococcus lactis*—and avelumab, a monoclonal antibody targeting PD-L1. GEN-001 is designed to enhance anti-tumor immunity through modulation of the gut microbiota, promoting dendritic cell activation, improved antigen presentation, and inflammatory cytokine production. Avelumab acts as a checkpoint inhibitor by blocking the interaction of PD-L1 with PD-1 on T-cells, restoring anti-tumor immune responses. This combination is in phase 2 development for the treatment of patients with PD-L1–positive advanced gastric or gastroesophageal junction (GEJ) adenocarcinoma, showing promising anti-tumor activity and a manageable safety profile[1][5][7].
