Drug intelligence / Profile preview
GEN-003 is a therapeutic protein subunit vaccine developed by Genocea Biosciences for the treatment of genital herpes simplex virus type 2 (HSV-2) infection. The vaccine is composed of two recombinant HSV-2 antigens: a transmembrane deletion mutant of glycoprotein D (gD2ΔTMR) and a fragment of infected cell protein 4 (ICP4.2). These antigens are formulated with Novavax's Matrix-M2 adjuvant, a saponin-derived adjuvant designed to enhance both B cell (antibody) and T cell immune responses. The vaccine aims to reduce viral shedding and the frequency of recurrent genital lesions in individuals with established HSV-2 infection. Although GEN-003 demonstrated statistically significant reductions in viral shedding and lesion days in Phase II clinical trials, Genocea discontinued its development in September 2017 to pivot its focus toward neoantigen cancer vaccines.
