genakumab + tislelizumab

Genakumab + tislelizumab is an investigational combination of two monoclonal antibody drugs under evaluation primarily for cancer and immune/inflammatory diseases. **Genakumab** is an interleukin-1 beta (IL-1B) monoclonal antibody developed as an anti-inflammatory agent and is analogous to other IL-1 inhibitors (such as canakinumab). **Tislelizumab** is a humanized IgG4 monoclonal antibody targeting programmed cell death protein 1 (PD-1), developed to block the interaction of PD-1 with its ligands PD-L1 and PD-L2 and thereby restore and enhance antitumor T-cell immune response. Tislelizumab is already approved for several cancers and is used as an immune checkpoint inhibitor, while genakumab has shown efficacy in phase 2/3 trials for gout, systemic juvenile idiopathic arthritis, and other inflammatory conditions. The combination is being tested in phase 1 trials for advanced solid tumors, aiming to modulate tumor inflammation through simultaneous blockade of innate and adaptive immune pathways, with potential additional anti-inflammatory benefits.
