Gepotidacin (GSK2140944) is a novel, first-in-class triazaacenaphthylene bacterial topoisomerase inhibitor developed by GSK. It has a unique dual-targeting mechanism of action that selectively interacts with two key bacterial enzymes: DNA gyrase and topoisomerase IV (type II topoisomerases), which are responsible for bacterial replication. This novel mechanism allows gepotidacin to retain activity against target pathogens that are resistant to other antimicrobial agents, including fluoroquinolones. The drug has demonstrated in vitro activity against a broad spectrum of bacterial pathogens, including methicillin-resistant Staphylococcus aureus (MRSA) and multidrug-resistant E. coli. It has been evaluated in both intravenous and oral formulations in clinical trials.
