Overview

GHRH + ghrelin is a combination of two endogenous peptide hormones—growth hormone-releasing hormone (GHRH) and ghrelin—administered together to synergistically stimulate the release of growth hormone (GH). GHRH acts primarily via the growth hormone-releasing hormone receptor on pituitary somatotrope cells, while ghrelin is an endogenous ligand for the growth hormone secretagogue receptor type 1a (GHS-R1a). When administered in combination, these peptides produce a greater GH response than either alone due to their synergistic interaction at both hypothalamic and pituitary levels. This synergy has been demonstrated in humans and animal models, with potential applications in diagnostic testing for GH deficiency and possibly therapeutic use where enhanced GH secretion is desired[1][2][3][6]. The mechanism involves both direct stimulation of their respective receptors as well as possible cross-talk between signaling pathways, including evidence that GHRH can interact with the ghrelin receptor[3][6].
