Overview

GI-102 + eribulin is a combination therapy being investigated for the treatment of advanced and/or metastatic solid tumors, including breast cancer and melanoma. **GI-102** is a novel bi-specific Fc fusion protein combining the CD80 ectodomain and an engineered interleukin-2 (IL-2) variant connected via a human IgG4 Fc. This construct targets both **CD80** (to block CTLA-4 on regulatory T cells) and **IL2Rβγ** (stimulating NK and CD8 T cell proliferation, while carrying reduced affinity for IL2Rα and thus less effect on regulatory T cells)[3]. **Eribulin** is a microtubule dynamics inhibitor—specifically, a synthetic analog of halichondrin B derived from marine sponges—that acts as a mitotic inhibitor and induces cell cycle arrest at G2/M by preventing microtubule polymerization, ultimately leading to apoptosis[4][2][6]. The combination is currently in phase 1/2 clinical development to assess safety, tolerability, pharmacokinetics, and preliminary therapeutic activity[3].
